  【摘 要】目的:探讨小鼠腹腔注射植物凝集素(PHA)后血液、脾脏、骨髓中的MDSC 数量的变化,明确PHA 对小鼠血液、脾脏、骨髓MDSC 的作用. 方法:小鼠腹腔注射PHA 后,提取小鼠血液、脾脏及骨髓进行细胞裂解,标记抗体后采用流式细胞仪进行检测和分析. 结果:PHA 处理组小鼠的骨髓、脾 脏、外周血中MDSC 百分比明显比生理盐水组高,骨髓中生理盐水组双阳性平均为36.6%,而PHA 组双阳性达到56.4%,差异有统计学意义(P<0.05). 脾脏中,生理盐水组双阳性平均为3.5%,然而植物凝集素组双阳性能达到11.5%左右,差异有统计学意义(P<0.05). 外周血中,PHA 处理组MDSC 百分比明显升高,对照组MDSC 平均百分比占4.6%,而PHA 组MDSC 平均百分比占19.8%,差异有统计学差异(P <0.05).结论:植物凝集素腹腔刺激小鼠后,可促使小鼠血液、脾脏、骨髓中MDSC 浓度的上调.

  Study on the effect of phytohaemagglutininon upregulating MDSCs of immune organ in mice

  【Abstract】AIM: To investigate the changes in the number of MDSCs in blood, spleen and bone marrow after intraperitoneal injection of phytohaemagglutinin (PHA) in mice, and confirm the effects of PHA on MDSC in blood, spleen and bone marrow.METHODS: After intraperitoneal injection of PHA in mice, the red cell extracted from the blood, spleen and bone marrow was lysised, and MDSCs altering of labeled antibody was detected and analysed using flow cytometry. RESULTS: Percentage of MDSCs treated by Phytohaemagglutinin was significantly higher than that of the control group in bone marrow, spleen, peripheral blood.More detail, MDSCs in control group was 36.6% in bone marrow,while MDSCs in Phytohaemagglutinin group reached 56.4%, and the difference was statistically significant (P<0.05). Average value of MDSCs in spleen was 3.5% in control group, however that of the Phytohaemagglutinin group was 11.5%, and the difference was statistically significant (P<0.05). Similarly, it is difference in peripheral blood (4.6% vs 19.8%). CONCLUSION: PHA can promote up⁃regulation of MDSC in blood, spleen and bone marrow in mice.
